Interferon gamma (IFNg) production is associated to disease parameters in TLR9-induced secondary hemophagocytic lymphohistiocytosis (sHLH) in mice

چکیده
Objectives We evaluated whether the neutralization of IFNg in murin sHLH and fulminant sHLH affected the disease features and we investigated whether treatment with an anti-IFNg mAb affected the clinical and laboratory features in murine models of sHLH and fulminant sHLH; we also exploited the in vivo principle that, in the presence of an anti-IFNg antibody, circulating IFNg bound to the antibody is incorporated in a complex leading to accumulation of the cytokine in serum, therefore allowing the quantification of IFNg production.

Interferon gamma (IFNg) drives disease in the TLR9-mediated cytokine storm syndrome in mice
The Cytokine Storm Syndrome (CSS) is characterized by an overwhelming activation of immune cells observed in life-threatening disorders such as familial hemophagocytic lymphohistiocytosis (fHLH) and secondary (s) HLH/ macrophage activation syndrome (MAS) as well as during serious infection. However, it is not known if the CSS can be attributed to a single cytokine. Increased blood levels of int...
متن کاملSuccessful Treatment of Scrub Typhus - Associated
Scrub typhus is caused by Orientia tsutsugamushi. Any delay in diagnosis can result in delayed treatment and severe complications, including secondary hemophagocytic lymphohistiocytosis, which is rare but potentially fatal. In this paper, the authors present 3 cases of secondary hemophagocytic lymphohistiocytosis associated with scrub typhus, successfully treated with chloramphenicol without ad...
متن کاملAnti interferon-gamma (IFNγ) monoclonal antibody treatment in a patient carrying an NLRC4 mutation and severe hemophagocytic lymphohistiocytosis
Background A growing body of evidence, in animals and humans, suggest that IFNg plays a pathogenic role in primary HLH (pHLH) and in secondary HLH, including macrophage activation syndrome. A pilot study in pHLH with NI-0501, an anti-IFNg monoclonal antibody, is ongoing. Mutations in NLRC4 have been recently reported to cause recurrent MAS and an increased production of IL-18, a cytokine known ...
متن کاملThe influence of various therapeutic regimens on early clinical and laboratory response and outcome of children with secondary hemophagocytic lymphohistiocytosis
Introduction Secondary hemophagocytic lymphohistiocytosis (sHLH) is a life-threatening syndrome of severe hyperinflammation which is often triggered by infection or autoimmune disease (macrophage activation syndrome - MAS). The aim of our study was to assess the frequency of sHLH/MAS in children treated in our institution and to compare the effectiveness of various therapeutic interventions. ...
متن کاملTreatment of MAS and HLH
Macrophage activation syndrome (MAS) is a lifethreatening complication of inflammatory disease, occurring secondary to a complex interplay of genetic factors, drugs, infectious agents and immunological anomalies. Early identification and aggressive treatment are mandatory to prevent fatal evolution. Precipitating factors should be looked for and eventually removed such as infections (leishmania...
